Autophagic Dysfunction
In this experiment, we wanted to see whether inclusion formation in dopaminergic neurons was linked to changes in autophagy. Under the dual treatment with α-synuclein fibrils and immune challenge, LC3B formed bright puncta that clustered around the developing inclusions, showing that autophagic vesicles were being recruited to these sites.
These findings revealed that autophagy is actively engaged at the sites of Lewy body–like inclusions, but instead of clearing fibrils, the machinery appears to become sequestered, potentially contributing to the buildup of pathology.
